ABSTRACT |
The effect of 2,4-D, IAA, NAA and kinetin on the growth of blade, midrib, sporophyll and holdfast explants in Undaria pinnatifida of Wan-do were investigated from February to March 1991. Each plants was cultured for 4 weeks on PESI medium supplemented with 2,4-D, IAA, NAA and kinetin, from 10 super(-4) M to 10 super(-8) M concentrations, respectively. In the blade explants, the most effective concentrations on the growth was commonly 10 super(-6) M in 2,4-D, IAA, NAA and kinetin. Also growth was promoted at lower concentrations (10 super(-6) M - 10 super(-7) M) and, in general, inhibited at a high concentration (10 super(-4) M). In the midrib explants treated with 2,4-D, IAA and NAA, growth was not promoted. On the contrary, in those treated with 10 super(-5) M - 10 super(-7) M of kinetin, the growth was promoted. The most effective concentration on the growth was 10 super(-7) M of Kinetin. In the sporophyll explants, the most effective concentrations on the growth was commonly 10 super(-5) M in 2,4-D, IAA and NAA. But treated with kinetin, growth was not promoted. In the holdfast explants, the most effective concentrations on the growth was 10 super(-5)M in 2,4-D, 10 super(-5) M in IAA, 10 super(-6) M in NAA and 10 super(-6) M in kinetin. At a high concentration (10 super(-4)) of four growth regulators, in general, the growth in holdfast explants was inhibited while at lower concentrations (10 super(-5) M - 10 super(-6) M), the growth was promoted. As a rule, growth of various explants was promoted at low concentrations (10 super(-6) M - 10 super(-7) M), whereas growth was inhibited at a high concentration (10 super(-4) M). Growth of various explants treated with 2,4-D, IAA, NAA and kinetin was in order of blade>holdfast>sporophyll>midrib.
