Characteristics of Triacylglycerols and Fatty Acids from Detonula sp. in Culture

Detonula sp. in Complete Medium (CM) for 4 weeks were cultivated in Nitrogen Absence Medium (NAM) for 3 weeks (state A) and transferred to CM and cultivated for 3 weeks (state B). Their triacylglycerols (TAG) and fatty acids were extracted. The ratio of the amount between triacylglycerols and total lipids (TL) after 4 weeks of the culture was 8.9% in CM (TAG: 2.5mg·g super(-1) dry wt, TL: 28.1mg·g super(-1) dry wt) but 33% in NAM (TAG: 24.5 mg·g super(-1) dry wt, TL: 73.7 mg·g super(-1) dry wt). The amounts of TL and TAG from Detonula sp. in the state A (TL: 70 mg·g super(-1) dry wt, TAG: 34 mg·g super(-1) dry wt were abruptly decreased in state V (TL: 1.5 mg·g super(-1) dry wt, TAG: 0.7 mg·g super(-1) dry wt). The dominant fatty acids were palmitic (C16:0)(27%), palmitoleic (C16:1)(46%) and oleic acid (C18:1)(7.6%) in CM and C16:0 (29%), C16:1 (45%) and linoleic acid (C18:2)(6.7%)in NAM. The C16 of Detonula sp. was abundant but C20 was diminished in CM and NAM for the duration of the culture.
